Figure Lengend Snippet: Pro-inflammatory and anti-inflammatory markers were assessed by immunoblot in 18-month old Cre+ChAT+ and control mice. Cortical lysates (25 µg) from male ( a ) and female ( b ) mice were separated using 8 or 12% SDS-PAGE gels, then transferred to PVDF membranes and probed with antibodies against target proteins. Lanes 1–4 represent Cre-ChAT+ mice and lanes 5–8 represent Cre+ChAT+ mice. The 82-kDa ChAT expressing mice exhibit higher levels of markers associated with neuroprotection and anti-inflammation and lower levels of pro-inflammatory markers. Representative blots show both male ( a ) and female ( b ) Cre+ChAT+ mice exhibit lower levels of C3 (ii), C1q (iii), Il1 beta (iv), Il6 (v), iNOS (vii) and Cd86 (x) and higher levels of Il4 (vi), Arg1 (viii) and Cd206 (xi) compared to control mice, without significant differences in the level of the microglial marker, Iba1 (i) between the two genotypes. Additionally, ratios of iNOS/Arg1 (ix) and Cd86/Cd206 (xii) are significantly lower in Cre+ChAT+ mice. Immunopositive bands from 4 biological replicates/sex/genotype were analyzed by densitometry and all samples were normalized to the loading control beta-actin. Data are shown as mean ± SD and statistical analysis was performed using unpaired Student’s t -test with asterisks denoting *p ≤ 0.05 relative to control.
